AbstractOne of the new challenges of single particle reconstruction is protein mapping inside the particle structure with the help of fluorescence imaging. To this end, we present a method for reconstruction of single particles from widefield data. We propose a joint deconvolution and multiview reconstruction approach dedicated to 3D fluorescence imaging. Our method is able to handle a large amount of 3D observations with a computationally efficient augmented-Lagrangian based optimization. Experimental results on synthetic data attest the performance gain yielded by our method over standard tomography and averaging approaches.
